pBV220載體是我國預(yù)防醫(yī)學(xué)科學(xué)院病毒研究所自行構(gòu)建的大腸桿菌溫控表達(dá)載體,目前仍在廣泛使用.
pBV220質(zhì)粒的SD sequence(用于結(jié)合原核生物核糖體的序列)后緊跟多克隆酶切位點(diǎn),便于插入帶起始ATG的外源基因,可表達(dá)非融合蛋白。
載體含有強(qiáng)的轉(zhuǎn)錄終止子可 防止出現(xiàn)"通讀"現(xiàn)象,有利于質(zhì)粒-宿主系統(tǒng)的穩(wěn)定。
載體pBV220為3.7Kb,有利于增加其拷貝數(shù)及容量。pBV220含有PL啟動子和編碼對該啟動子具有抑制作用而又對溫度敏感的cI蛋白基因cIts857調(diào)控基因cI,因此可用溫度對插入其中的外源基因的轉(zhuǎn)錄進(jìn)行調(diào)控。
溫控型原核表達(dá)載體,高拷貝數(shù),大容量,強(qiáng)啟動子,強(qiáng)轉(zhuǎn)錄終止子,可在任何受體菌中誘導(dǎo)表達(dá)。
pBV220載體是λPL/PR-cI857溫度敏感系統(tǒng)表達(dá)載體,能夠在42 ℃表達(dá)非融合的目的蛋白。
在利用pBV220載體進(jìn)行質(zhì)粒構(gòu)建的時候,需要加入ATG起始密碼子。
